Abstract | Garcinia mangostana Linn. (Clusiaceae) is a tropical evergreen tree in Southeast Asia, and its fruits, mangosteen, are known as “Queen of Fruit” in Thailand. The pericarps of G. mangostana have long been valued as an indigenous medicine for treating catarrh, cystitis, diarrhea, dysentery, fever, skin infections, wounds, etc. This dissertation focused on the chemical and bioactive investigation of G. mangostana. Phytochemical investigation was carried out on the chloroform fraction of the EtOH extract of the pericarps of G. mangostana. A total of 27 compounds were isolated with comprehensive chromatography technologies, and identified with various spectroscopic methods. Among them, seven compounds were new, including three dimeric xanthones, namely cratoxyxanthone B-D (1-3), and four monomeric xanthones, namely laterixanthone B-E (4-7). The structures of new compounds were elucidated by 1D and 2D-NMR spectra and other spectroscopic studies. 20 known compounds were identified by 1D-NMR and MS data. Among them, 7-O-methylgarcinone E and trapezifolixanthone were found from G. mangostana for the first time. Bioactive constituents screening found that many xanthones from G. mangostana had potent anti-oxidative, anti-inflammatory and antitumor activities. In current study, anti-oxidative and anti-inflammatory activities of ten major xanthones, and antitumor effect of twenty-five xanthones were tested. Among them, γ-mangostin showed the most significant anti-oxidative and anti-inflammatory activities. Garcinone E and trapezifolixanthone exhibited the most significant antitumor activity on A549 lung cancer cell and HEY ovarian cancer cell, respectively. In the current thesis, 7 novel xanthones were identified from G. mangostana; and the chemical principles for its anti-oxidative, anti-inflammatory and antitumor activities were achieved. These findings supply the first key step for the further development of G. mangostana and its constituents as drugs or healthy food. |
