AbstractThe development of female mammary glands usually starts during puberty and is dramatically enhanced in pregnancy, as the result of accelerated proliferation and differentiation of mammary stem cells (MaSCs) in response to sexual hormones stimulation. Although the hierarchy of mammary epithelia has been studied by marker analysis and cell fate tracing, the molecular characters of MaSCs remain elusive, partly due to lack of exclusive markers for MaSCs and undiscovered transcriptome complexity via conventionally ensemble measurements of gene expression in bulk of cells. Hence, in this study we performed comprehensive and high-resolution transcriptome analysis of 239 single mammary basal and luminal epithelial cells from virgin and pregnant mice. Based on their respective gene expression profiles, we unbiasedly classified the single mammary cells into seven distinct subgroups with typical markers and enriched biological functions, indicating their locations and properties in mammary hierarchy. Moreover, pseudo-temporally reconstructed mammary epithelia developmental trajectory reveals unidirectional and continuous differentiation of MaSCs in virgin state but bidirectional differentiation into basal and luminal lineages separately in pregnant mice. In sum, this study provides significant insights into heterogeneity and hierarchy of mouse mammary epithelia.
