Norditerpenoids from Podocarpus nagi and their anti-cancer activity

AbstractPodocarpus (Podocarpaceae) genus plants are widely distributed in South of China, of which about 30 kinds of roots, stems, leaves, flowers are used as traditional medicines for treatment of fevers, asthma, coughs, cholera, distemper, chest complaints and venereal diseases. Norditerpenes and totarane-type diterpenoids from Podocarpus are known to have cytotoxic activities against several forms of cancer. To searching novel anti-cancer agents, the seeds of Podocarpus nagi, collected from Yunnan China, were systematically isolated. ??? norditepenoids were identified from the ethyl acetate and n-butanol fractions, including parts The totarane-type diterpenoids are isolated from of and are proved to have remarkable anti-tumor activity by virtue of in vitro anti-tumor activity tests. The air-dried seeds of P. nagai were ground into powder and extracted with 95% ethanol for three times. After evaporation of the collected percolate, the crude extract was suspended in H2O and extracted with chloroform, ethyl acetate and n-butanol, successively. The ethyl acetate and n-butanol fractions were subjected to chromatography over silica gel and eluted with petroleum ether–acetone (19:1 to 0:1, v/v), respectively. Each subfraction was further separated over MCI gel, Sephadex LX-20 gel, ODS gel, and preparative HPLC, to afford nagilactones A, B, D, E and related novel structural derivatives.Phytochemical investigation on Podocarpus nagi led to the isolation of several new diterpenoids. These were characterized based on 2D NMR, IR, UV, and MS spectroscopic analysis and their absolute configurations were determined by single crystal X-ray studies, as well as by 1H NMR spectroscopic analysis for the corresponding chiral derivatives. The totarane-type diterpenoids show potent anti-tumor activities on different cancer cell lines, including lung cancer (A549), ovarian cancer (A2780), liver cancer (HepG2), breast cancer (MCF-7), stomach cancer (SGC7901) and melanoma (A375). These substance are applied to treat different kinds of cancer cells (IC50=0.53±0.37).
