Abstract | As a hybrid imaging technique, photoacoustic imaging (PAI) can provide multiscale morphological information of tissues and can recover the spatial distribution of interesting chromophores such as hemoglobin within tissues using multi-spectral PAI (MSPAI). Herein, we develop a contrast agent that can high-effectively combine multiscale PAI with MSPAI for a more comprehensive characterization of a complex biological tissues. In this study, we developed a novel PIID-DTBT based semiconducting polymer dots (Pdots) that have broad and strong optical absorption in the visible-light region (500 nm - 700 nm). Gold nanoparticles (GNPs) and gold nanorods (GNRs) that have been verified as an excellent PA contrast agent were compared with Pdots based on the multiscale PAI system. Both ex vivo and in vivo experiment demonstrated Pdots have a better PA conversion efficiency at 532 nm than GNPs and similar PA performance with GNRs at 700 nm at the same mass concentration. Photostability tests and toxicity tests demonstrated Pdots are photostable and biocompatible. More importantly, in vivo MSPAI experiment indicated Pdots have great PA performance than the blood and can be accurately extracted from the background of vascular-rich tissues. Our work demonstrates the great potential of Pdots as a highly effective contrast agent for precise localization of lesions relative to the blood vessels based on multiscale PAI and MSPAI. |
