AbstractObesity has reached epidemic proportion worldwide. Adipocyte dysfunction is a major cause of obesity. Screening for the efficacy on lipid accumulation in 3T3-L1 cells is an effective tool for the identification of new anti-diabetic compounds.1 The current anti-obesity drugs have severe side-effects. Natural products from commonly used fruits or vegetable may be an excellent alternative for developing future effective, safe anti-obesity drugs.2 The fruit mangosteen (Garcinia mangostana Linn.), called “Queen of Fruit”, is widely used in Southeast Asia for treatment of variety of medical conditions. We observed the crude extract from G. mangostana inhibited adipocyte differentiation in vitro, as evidenced by a decrease in cellular lipid accumulation. Thus, we hypothesize that chemicals from G. mangostana inhibit adipogenesis, which could potentially be used as a novel anti-obesity agent. Under the guideline of cell-based adipogenesis assay, 36 xanthones were isolated and identified from the pericarps of G. mangostana, including six new compounds (1-6). Compound 1 significantly reduced lipid accumulation, suppressed reactive oxygen species production, and increased basal and insulin-stimulated glucose uptake in adipocytes.
