Abstract | Sphingolipids are relatively minor membrane component serving as a structural molecule by forming micro domains on the plasma membrane. The metabolites of sphingolipids such as ceramide, sphingosine and sphingosine 1-phosphate (S1P), serving as signaling molecules with an extremely small amount, play a crucial role in modulating cell signaling. It is indicated that their signaling systems have been involved in diverse biological processes, including organ morphogenesis. Likewise, perturbations of sphingolipid signaling molecules have been associated with organ morphogenesis. Unlike growth factors and cytokines, the lipid mediator is not coded directly by the genome, thus the physiological activity of S1P is regulated by the spatiotemporal control of the S1P metabolizing enzymes, transporters and receptors. S1P can either be secreted outside cells acting with its receptors, or in some cases can function as an intracellular second messenger independent of S1P receptors. These observations suggest that elucidation on the metabolism and mechanism of S1P signaling can contribute to understanding organ morphogenesis defects. This project aims at revealing the mechanism of S1P function on organ morphogenesis. |
