Abstract | Cucurbit[n]urils (CB[n]), an emerging family of macrocyclic molecules, consist of n glycoluril units connected by 2n methylene groups, and they possess not only a hydrophobic internal cavity that can encapsulate various lipophilic guest molecules, but also the two carbonyl-fringed portals that are good cation-receptors. The chemical and biological activity of a guest molecule is often altered upon encapsulation by CB[n]. During the past several years, a number of reports (including ours) regarding the encapsulation of bioactive molecules including several important phytochemicals have demonstrated that CB[n] encapsulation often leads to improved solubility, chemical stability, and therapeutic efficacy as well as reduced side-effects. Our research group has been actively investigating CB[n]’s biocompatibility and its complexation with a variety of biomedically important phytochemicals during the past a couple of years, from both phytomedicine and pharmaceutical sciences perspectives. For instance, we have discovered that supramolecular encapsulation of several phytochemicals by CB[n] may help alleviate the toxic side-effects of these molecules, whereas their therapeutic efficacy is not compromised (in some cases, the therapeutic activities are even improved). We are also among the first to investigate CB[n]’s potential as a reversal agent for toxic phytochemicals and synthetic agents. In this presentation, we will discuss about our recent progress of these efforts. |
