Gold nanorods with noncovalently tailorable surface for image-guided chemo-photothermal cancer therapy

AbstractGold nanorods (Au NRs) have drawn significant attentions in recent years due to their promising applications in plasmon-enhanced spectroscopies, sensing, bioimaging, photothermal therapeutics and drug delivery. However, Au NRs surface functionalization strategies are often tedious and the commonly used surfactant CTAB often exhibit serious cytotoxicity. A facile surface functionalization method is needed to endow Au NRs with excellent biocompatibility and the capability for targeted payload delivery, and photothermal therapy. Herein we present facile, non-covalently tailorable surface functionalization of Au NRs for multiple functional, targeted chemo-photothermal therapy and theranostics. In this work, The surface of Au NRs were covalently modified with mono-alloxyCB[7] (AO1CB[7]), which allowed non-covalent tailorable conjugation with amantadine (Ada)-tagged functional molecules via strong CB[7]-Ada host-guest interactions. Ada-folate (Ada-FA), and chemotherapeutic agent oxaliplatin (OX) were respectively used as the model targeting molecule and chemotherapeutic drug to get modularly attached onto Au NRs surface, to enable targeted payload delivery and chemo-photothermal therapy. Particularly, Ada-FA and OX loaded Au NRs specifically targeted FA-receptor overexpressed cancer cells and released OX via competitive replacement by spermine (overexpressed by cancer cells). Upon cellular uptake, irradiation of Au NRs by external NIR laser (808 nm) led to cellular hyperthermia, and the temperature increase may also accelerate OX release. Moreover, these Au NRs showed great potential in computed tomography (CT) imaging, optical coherence tomography (OCT) imaging and photoacoustic (PA) imaging, affording a promising theranostic nanoplatform for imaging-guided cancer chemo-photothermal therapy.
