Alisol B 23-acetate induces apoptosis and G1 phase arrest, and inhibits migration and invasion on ovarian cancer cells

AbstractOvarian cancer is the first leading cause of death among gynecologic malignancies worldwide. Discovery of new chemotherapeutic drugs is still imperative for the improvement of the survival rate. This study aims to investigate the anticancer potential of alisol B 23-acetate (23B), a protostane-type triterpene isolated from the Alismatis Rhizoma, on ovarian cancer cells including HEY, A2780, and A2780/TR (paclitaxel-resistant A2780). MTT and Hoechst staining assays were performed to evaluate the cell cytotoxicity of 23B, along with flow cytometry and Western blotting for apoptosis and cell cycle analysis. Wound healing and transwell assays were performed to investigate the effect of 23B on cell migration and invasion. Meanwhile, the autophagy related proteins were also detected using Western blotting. The results showed that 23B inhibited cell viability and induced accumulation of the Sub-G1 phase in the three cell lines in a concentration dependent manner. The protein levels of cleaved poly ADP-ribose polymerase (PARP) and Bax/Bcl-2 were up-regulated after treatment with 23B. It could block the cell cycle progressions in G1 phase through down-regulating the protein levels of CDK4, CDK6, and cyclin D1. Wound healing and transwell assays showed that 23B suppressed the migration and invasion of HEY cells at low concentration. Moreover, it could up-regulate the protein level of LC3-II and p62 of the three cell lines in time- and concentration- dependent manners, indicated that it may inhibit autophagic flux by the lated stage. In conclusion, 23B possessed anti-proliferation and anti-metastasis activities on ovarian cancer cells.
