Abstract | Lung cancer is one of the most formidable health threats all around the world. In the latest WHO cancer statistics report released in 2014, lung cancer was the notorious cause of death which accounted for 23.6% and 14.7% cancer deaths in men (11 million people, No.1 cause of death) and female (0.5 million people, No.2 cause of death, followed by breast cancer) respectively. In 2015, Macau Cancer Registry also indicated that lung cancer was the most leading cancer disease in both morbidity and mortality. In order to understand the tumorigenesis mechanisms of lung cancer thoroughly, the somatic mutation-based genome studies become the research mainstream to date. The activation of oncogenes and inactivation of tumor suppressor genes may be triggered by somatic mutations, which allow deleterious cells keep proliferating with selective growth advantages during lung cancer development. We hypothesize that somatic mutation profiles of different subgroups of lung cancer patients are different. In the present study, next-generation DNA sequencing data of non-small lung cancer tumors in 2805 cases were retrieved from public cancer genome database cBioPortal. 218 significantly mutated genes (SMGs) were observed with TP53, KRAS EGFR, STK11, KEAP1 being the top five SMGs. We compared profiles of SMG among different subgroups of lung cancer patients. Somatic mutation rates of some genes were significantly different among patient subgroups of squamous lung cancer (e.g., TP53 and EGFR in smoker and non-smoker, chi-square test). Our results suggested that these SMGs might be potential lung cancer drug targets. |
