Abstract | Protien glutaminase (PG) from Chryseobacterium proteolyticum is a safe and effective deamidating enzyme which can improve solubility and emulsification ability of food proteins through deamidation modification. The use of PG can decrease the abuse of food additives. Despite PG can deamidate many food proteins, the limited catalytic efficiency has restricted the utilization of PG in the food industry. Correlation between the protein structure and catalytic activity can provide insights on how to enhance the catalytic efficiency of PG through protein engineering. In this study, we identified a PG homologue in GenBank sequence database using Protein Blast. Subsequently, we produced a recombinant pro-protein of the PG homologue (proPG homologue), and obtained its high resolution crystal structure. The overall structure of the proPG homologue showed high homology to the crystal structure of the pro-protein of PG, and shared the similar catalytic triad (Cys-His-Asp). However slightly differences were observed in the active center. In conclusion, we have discovered a protein which shares high degrees of sequence homology and structural homology with PG. Experiments on enzymatic characterization of the PG homologue are been carried out in order to establish the relationship between the protein structure and catalytic activity of PG. |
