BDNF Induce the expression of Synapsin I in Primary Cultured Cortical Neurons

AbstractNormal synapse struture and functions are essential for learning and remembering.BDNF is a major neurotrophic factor with an important role in the development of synapse formation. To study the role of BDNF in the formation of synapse, we investigated the effect of BDNF on the expression of synapse I in primary cultured cortical neurons. The effect is time- and concentration-dependent with the maximal effect of BDNF at 100ng/ml and 6 h respectively. Western blot with phospho-antibodies for various kinases showed that BDNF activated the MAPK and the PI3K pathways in these culture neurons. Blockage of the activation of PI3K pathway with PI3K specific inhibitor LY294002 attenuated the expression of synapse I induced by BDNF, while less effect was seen for the MAPK pathway inhibitoe PD98059. These results put together indicate that BDNF is able to induce the expression of synapse I in primary neurons via the PI3K pathway
