α-Mangostin protects against age-associated metabolic disorders targeting adipose tissue inflammation

AbstractObesity and its related metabolic disorders are highly prevalent in the elderly individuals. Low-grade chronic adipose tissue inflammation contributes to the onset and development of age-related insulin resistance and type 2 diabetes. α-Mangostin (α-Man) is an anti-inflammatory xanthone from Garcinia mangostana. The present study was designed to verify the hypothesis that α-Man could alleviate adipose tissue inflammation to mitigate age-related metabolic disorders. α-Man suppressed lipopolysaccharide (LPS)-induced inflammatory responses in RAW264.7 macrophages, and tumor necrosis factor α-mediated inflammation in 3T3-L1 adipocytes, through inhibiting the mitogen-activated protein kinases and nuclear factor-κB activation. In cell co-culture system, α-Man blocked RAW264.7 macrophages migration towards 3T3-L1 adipocytes. Furthermore, α-Man prevented mice from LPS-induced acute adipose tissue inflammation by reducing pro-inflammatory cytokines and chemokines expressions and inhibiting macrophages migration into adipose tissue and the pro-inflammatory polarization of macrophages. In a cohort of young (3-month) and old (18‒20-month) mice, α-Man obviously mitigated age-associated adiposity, hyperlipidemia and insulin resistance. α-Man alleviated age-related adipose tissue inflammation by reducing macrophage infiltration and pro-inflammatory polarization in adipose tissue. Moreover, α-Man reduced liver lipotoxicity through suppressing the secretion of microRNA-155-5p from macrophages. The above results demonstrated that α-Man might represent a novel candidate to treat aging-related metabolic disorders targeting adipose tissue inflammation.
