Abstract | Aim: To establish a HPLC-FLD method for determining the concentration of aesculin in rabbit plasma. Methods: The sample was separated with a C column (150 mm x 4.6 mm, 5 μm) at 30°C with the mobile phase of methanol and water (74:26); A fluorescence detector was used (λex = 230 nm λem = 460 nm); The flow rate was 1 ml·min. Results: The retention time of aesculin was (3.620 ± 0.05) min; A good linear relationship of aesculin in the plasma samples at the concentration between 0.1 mg·L and 100 mg·L was observed (r = 0.997); The mean absolute recovery was 74.0%-82.6% and the lowest detective limit was 0.05 ng; Both the intra-day and inter-day precisions were less than 9.0%. After a single intravenous injection of aesculin (5 mg·kg) in rabbits, the T and T were (0.196 ± 0.008)·h and (6.70 ± 0.645)· h. Conclusions: The method was simple, sensitive and credible and could be used for the pharmacokinetic study of aesculin. |
