Abstract

本发明公开了表达抗CD22嵌合抗原受体和PD‑L1阻断蛋白的细胞的制备方法、表达载体及应用,涉及医药生物技术领域。表达载体插入有编码抗CD22嵌合抗原受体的第一核酸和编码PD‑L1阻断蛋白的第二核酸。利用CD22靶标和PD‑L1靶标,通过靶向CD22的抗体或其片段,本发明的T细胞可高效、特异性地靶向表达抗原CD22的肿瘤细胞;PD‑L1阻断蛋白可以与内源表达的PD‑1竞争结合靶细胞上的PD‑L1,从而阻断PD‑1/PD‑L1信号通路,延长了本发明上述T细胞的作用时间,提高了肿瘤杀伤作用,由此缓解了实体瘤对CAR T细胞疗法的免疫耐受性的问题。
