Abstract | Scorpion is one of the virtually untapped sources of new pharmaceutically active agents for the treatment of diseases. In the last few decades, scorpion venom has been proved to have rich sources of bioactive peptides, which play an important role in defence or prey in the complex natural environment. Our previous studies indicated that many of the venom-based bioactive peptides demonstrated with high pharmacological diversities, which have the ability to kill bacterial, fungi and even cancer cells. In this study, we developed a high-throughput approach to identify novel peptides from scorpion venoms by using RNA-seq next generation sequencing, and de novo peptidomics approach. Meanwhile, we performed the traditional method and compared the results with the new approach. Those venom peptides confirmed from the two approaches were screened by a series of pharmacology functional assay to evaluate their therapeutic potential. So far, two novel peptides have been identified from the venom of the scorpion Androctonus aeneas, AaeAP1 and AaeAP2, which have the significant anticancer activity on four human cancer cell lines, including H460, MB435s, MCF-7 and PC3. In conclusion, this project provides a new approach to identify bioactive peptides from venomous animals in a high-throughput manner, and paves a new insight of using venom-based peptide for the treatment of cancer. |
