Association of TAZ, AXL and CTGF expression with clinicopathological parameters in colon cancer

AbstractColon cancer accounts for a large proportion of cancer related morbidity worldwide. Genetic analysis and stratification of patients based on survival can identify genetic signatures that may be used for prognostic or treatment planning purposes. We have previously shown that the TAZ-AXL-CTGF mRNA signature could predict patient survival in two independent colon cancer datasets. However, limited clinicopathological data were available from these two datasets. Recently, a large colon cancer dataset was published in the GEO database (totaling 566 patients) that contains data such as tumor stage, tumor location, MMR status, KRas status, Braf status and p53 status. The mRNA expression levels of TAZ, AXL and CTGF were extracted, and TAZ-AXL-CTGF signature was analyzed as in our previous report. The TAZ-AXL-CTGF signature was then correlated with available pathological parameters and survival. We found that a high level of TAZ-AXL-CTGF expression, defined by high level expression of all these three genes, was associated with higher pathological stage, deficiency in mismatch repair, colon cancer subtype C4 and BRAF mutation. Importantly, TAZ-AXL-CTGF overexpression was associated with shorter overall survival in patients with, TP53 mutation, colon cancer subtype C6, proficient MMR, wild type KRAS and BRAF status. Importantly, the prognostic value of TAZ-AXL-CTGF overexpression is independent with all clinicopathological parameters and mutation statuses. This report confirms our previous analysis suggesting that TAZ-AXL-CTGF mRNA signature is a potential prognostic indicator in colon cancer.
